Direct DNA sequencing will be a routine procedure in specialized clinical laboratories within 10 years-Editorial in Clin. Chem. 41/10, 1407-1409 (1995)

The major roadblock to realizing the above prediction is the cost of preparing clinical specimens for direct DNA sequencing. LoTemp™ HiFi® PCR has overcome this roadblock by eliminating the needs for DNA extraction and purification, thus opening the gateway to direct DNA sequencing for all clinical laboratories with a dedicated technical staff. The potential of product development is enormous. Currently, we offer the following to our customers:

(1) LoTemp™ HiFi® PCR ready-to-use Mix 1 mL x 2. Catalog #8802

Ready-to-use mix contains optimized HiFi® DNA polymerases, buffer, Mg++, dNTPs, stabilizers, and proprietary melting reagents for purified and unpurified DNA samples. Use 20 mL for each 25 mL final PCR volume, thermocycled at 40° - 85°C.  Its advantages are:

  • Produce more specific amplicons than any other heat-resistant enzymes

  • Nested PCR products used for direct automated DNA sequencing

  • Stable at room temperature for at least 4 weeks

  • Stable at 4°C  for at least 10 months  

  • Ideal for field PCR & large-scale PCR

  • Reduce DNA damage by LoTemp™ denaturing

  • Reduce in-house pipetting contamination

  • Just add primers and template

  • Do not need hot-starts

(2) β-globin PCR primer pair, optimized for specimen adequacy, amplicon

(3) PCR product transferring glass rods, calibrated, 100 rods.  Using thin glass rods to transfer PCR amplicons in nested PCR and for direct DNA sequencing reaction minimizes the risk of sample contamination.        Catalog #3324

        Items 4-15 are for preparation of PCR amplicons from clinical materials to perform direct automated Sanger DNA sequencing for genotype-specific signature sequence validation of the L1 gene of HPV, and for validation of the species-specific gonococcal opa genes, and for the species-specific cryptic plasmid DNA of Chlamydia trachomatis.
